Abstract: Heteroanionic solids have unique physicochemical features and offer chemists additional degrees of freedom for fine-tuning properties. However, access to multianion phase space has remained hindered by chemical bottlenecks that stem from mismatches in anion reactivity. While high-throughput computation and machine learning have emerged as powerful tools that can guide exploratory efforts, the prediction of truly novel inorganic solids remains elusive. This is especially true when approaching phase space with synthetically challenging combinations of elements, where little or no data exist. Furthermore, while order-disorder phenomena in mixed-anion systems is known to underpin correlated phenomena, these subtle crystallographic features are often overlooked and poorly understood. Accordingly, strategies that can circumvent these bottlenecks to access new multianion materials with atomic-scale precision are highly desirable. Here, I will describe chemistry our group has developed to access new families of oxychalcogenides using anion-based dimensional reduction, which is also used to engineer optoelectronic anisotropy in a controllable manner. In the second part of the talk, I will discuss how crystallographic analysis and hard-soft acid-base concepts can be used to predict the lattice positions that secondary anions will occupy in a range of single-anion chalcogenide and metal halide hosts. The analysis serves as a framework for generating libraries of sulfoselenide, selenotelluride, and sulfotelluride products with controllable degrees of anion ordering. I will also discuss how site-selective anion alloying can be used to break inversion symmetry, which has important implications for applications that rely on non-centrosymmetric crystal systems.

In the study of quantum matter, measurements have traditionally been viewed as a means of learning about a system. Measurements can, nevertheless, play a more active role—generating novel quantum phenomena that may be difficult or impossible to realize in measurement-free settings. As an interesting example, I will discuss how measurements can dramatically alter universal properties of quantum systems tuned to a phase transition. I will also highlight a path to experimental realization in analog quantum simulators based on Rydberg atom arrays. Finally, I will describe how these ideas inform optimization of quantum teleportation protocols against imperfections—establishing a long-term quantum science application of ‘measurement-altered quantum criticality’.

Abstract: Controlling highly reactive radical and high-valent metal intermediates is a central challenge in both biological catalysis and synthetic organometallic chemistry. My group integrates electron paramagnetic resonance (EPR) spectroscopy with cryogenic electron microscopy (cryo-EM) and computational approaches to directly observe and mechanistically characterize these fleeting species. In the first part of this talk, I will describe how substrate-triggered conformational dynamics activate and control radical chemistry in adenosylcobalamin-dependent mutases. Cryo-EM structures combined with EPR detection of paramagnetic intermediates, reveal that large-scale domain motions gate Co(III)–C bond homolysis and spatially confine the resulting 5′-deoxyadenosyl radical, providing a framework for how enzymes accelerate radical initiation while suppressing off-pathway damage. In the second part, I will show how EPR spectroscopy identifies and assigns transient high-valent copper and nickel intermediates in catalytic C–C bond formation. Capturing Cu(II), Cu(III), Ni(III), and formal Ni(IV) species under catalytic conditions provides direct evidence for radical-mediated oxidative addition pathways and unconventional high-valent redox cycles. Together, these studies demonstrate how combining structural, spectroscopic, and computational methods enables direct visualization of reactive intermediates and reveals unifying principles for controlling high-energy chemistry across biological and synthetic systems.

The paper discusses the history of Iranian-Jewish activism in Iranian socialist and communist circles. Following the 1941 Anglo-Soviet occupation of Iran, the political sphere opened to all parties and tendencies. The Comunist Tudeh Party was formed adn became the only party in Iran that allowed non-Muslims to become members. Iranian Jews supported the party for many reasons, ranging from its positions against antisemitism and Nazism to its work for social justice and advancement. Jewish activism remained significant until the 1979 Revolution. This talk, focused on Iranian Jewish activism, will examine the intersection of Iranian nationalism, Communism, and Zionism.

Department of Human Genetics Doctoral Candidate, Tabitha Banks-Tibbs, will defend the following dissertation on “In Situ Cryo-ET as a Marker of Mitochondrial Bioenergetics”

ABSTRACT:

Mitochondrial architecture varies dramatically based on cellular bioenergetics and is altered in disease states that impair the function of oxidative phosphorylation. This has been demonstrated by confocal light imaging and/or transmission electron microscopy (TEM). While these methods allow examination of altered mitochondrial morphology in two dimensions, they cannot resolve structural defects specific to individual respiratory complexes or higher-order supercomplex organization in three dimensions. Additionally, conventional TEM sample preparation (e.g., fixation, staining, dehydration) alters mitochondrial structure, making it challenging to disentangle disease-related changes.

Cryo-electron tomography (cryo-ET) utilizes TEM to acquire high-resolution, serial images of vitrified mitochondria in situ, allowing for the generation of three-dimensional images free from extrinsic manipulations that alter cellular architecture. Here, I use a cryo-ET pipeline that the Freyberg lab has developed to image vitrified fibroblasts derived from patients with genetically-defined mitochondrial diseases to: 1) identify alterations to mitochondrial ultrastructure in propionic acidemia, and 2) characterize the reversal of these changes as a result of a novel anaplerotic compound. Furthermore, I adapt the existing cryo-ET pipeline for use with T cells to investigate the effects of 1) physiological activation, and 2) the tumor microenvironment on mitochondrial ultrastructure. Notably, I have identified an increase in the intermembrane space (MIM-to-MOM distance) and cristae width via cryo-ET correlates to increased glycolytic flux in both patient-derived fibroblasts and T cells. These changes in mitochondrial ultrastructure are alleviated by the therapeutic agent in PA. I also show that tumor-infiltrating lymphocytes (TILs) exhibit an increase in ATP synthase nearest-neighbor difference, consistent with impaired oxidative phosphorylation.

How did hinterland households navigate life during the Mongol Empire? In Tarvagatai Valley, north central Mongolia, it was    through flexible multiresource pastoralism predicated on pastoralist animal management, small-scale agriculture, and household metallurgical production. This dissertation used a multimethod approach, combining paleoethnobotany, zooarchaeology, geophysical survey, and traditional excavation methods to holistically explore a unique household context from the Mongol period (13-14th century).

Rather than intensifying production for imperial markets or maintaining isolated traditional practices, households at Tsagaan Ereg grafted new economic activities onto existing pastoral lifeways without fundamentally reorganizing their livestock management. This household-scale perspective reveals how ordinary families in the empire's heartland actively shaped their own economic strategies, challenging elite historical narratives about imperial transformation.

Abstract:

Designing molecules and materials for engineering applications demands fast and reliable thermochemical property predictions. Semiempirical quantum mechanics (SEQM) methods and analytical force fields (e.g., ReaxFF) can screen candidates too numerous for ab initio quantum chemistry, but intrinsic empiricism limits their predictive power and obscures the physical drivers behind molecular behavior. Moreover, most traditional methods cannot decompose molecular energies into individual bond contributions, limiting the chemical insight available to guide molecular design. In this work, I developed a new family of methods called post-semiempirical quantum mechanics (post-SEQM), called BEBOP, ZPE-BOP, and QBOP, that achieve competitive accuracy while providing intramolecular energy decompositions that conventional methods cannot, enabling both property prediction and chemical interpretation at a cost bottlenecked only by a Hartree-Fock (HF) single point calculation.

BEBOP calculates total atomization energies physically partitioned across chemical bonds, achieving accuracy competitive with many Kohn-Sham Density Functional Theory methods for first row elements. ZPE-BOP computes zero-point vibrational energies from a single energy calculation, bypassing costly Hessian calculations that limit high throughput screening while preserving bond level energy decomposition. QBOP extends these vibrational descriptors to construct an analytical vibrational partition function and delivers thermal corrections (e.g., Gibbs free energy and heat capacity at constant volume) without requiring Hessians, providing essential thermodynamic quantities.

In Aim 1, I developed BEBOP models incorporating analytical σ/π bond order channels based on extended-Hückel theory, short-range repulsion interaction, and charge effects. I demonstrated that BEBOP descriptors encode essential chemical information by building a linear LASSO model that predicts deblocking temperatures in thermoplastic polyurethanes, validating their utility as molecular design tools for polymer engineering. In Aim 2, I developed ZPE-BOP and demonstrated its ability to capture vibrational energetics from bond level descriptors, offering a rapid route to vibrational properties for materials characterization. In Aim 3, I developed QBOP and established a framework for computing thermochemistry corrections entirely from vibrational bond energies, laying groundwork for thermodynamic property prediction in molecular and process design. Together, these post-SEQM methods establish a new paradigm for fast, interpretable, and physically grounded thermochemical predictions useful for generative chemical design applications.

Lunch Time Talk: Ákos Szegofi

Title: The misinformation-problem

Abstract:

How dangerous is misinformation? What effects does it have on beliefs and behavior, and how can we – should we? – defend against it? In recent years, two schools of thought have emerged to address these questions, that I will call “naivists” and “vigilantists.” The naivist school views misinformation as extremely dangerous, arguing that humans are overly gullible and/or lazy when evaluating communicated information, which makes them vulnerable to deception. The solution is to enhance people’s cognitive abilities and motivation. The vigilantist school holds that humans are epistemically vigilant, misinformation is not a new problem, and the solutions proposed by the naivist school have unintended consequences, such as decreasing trust in democratic processes and triggering widespread technology panic.

My research seeks to bridge these two schools by demonstrating that misinformation can be dangerous even if listeners are epistemically vigilant and update their beliefs rationally. I empirically test two, historically documented disinformation methods, then explore how modern communication environments enabled their widespread usage. I conclude by proposing structural solutions that focus on reshaping these environments, allowing listeners to trust more, instead of becoming cynical.

Chee-Hann Wu is an assistant professor faculty fellow in Theatre Studies at New York University. She received her Ph.D. in Drama and Theatre from the University of California, Irvine. Chee-Hann is drawn to the performance of, by, and with nonhumans, including but not limited to objects, puppets, ecology, and technology. Her current book project, provisionally titled The Unfinished Gesture: Puppetry, Memory, and the Becoming of Taiwan, considers puppetry a mediated means to narrate Taiwan’s cultural and sociopolitical development, colonial and postcolonial experiences, as well as Indigenous histories. Chee-Hann’s work has appeared in Puppetry International, Asian Theatre Journal, International Journal of Performance Arts and Digital Media, and other places. Her most recent research explores video games, VR, and artificial intelligence through the lens of theatre and performance. In addition to her scholarly work, Chee-Hann is the editor and a frequent contributor to Taiwan Insight, an online academic magazine of the University of Nottingham’s Taiwan Research Hub.

This exhibit, co-curated by students in the Kenneth P. Dietrich School of Arts & Sciences, showcases University Archives from the University of Pittsburgh Library System that document the transnational networks and economic class dynamics at play during the Room's development in the 1930s and 1940s. How did decisions made in Rome and Florence determine the Italian Room’s decorative style and use of materials? To what extent was the Italian immigrant and ethnic community in Pittsburgh involved in these decisions? And at what point did skilled manual labor intersect with philanthropic support and elite decision-makers to bring the Italian Room to completion? Abstract:

Per- and polyfluoroalkyl substances (PFAS) are among the most persistent and chemically resistant environmental contaminants, comprising thousands of diverse compounds used across industrial and consumer applications. While chemical redox and bioremediation strategies are being explored for PFAS removal, predictive computational frameworks capable of systematically evaluating structure-specific susceptibility to degradation and informing remediation approaches remain underdeveloped. This dissertation addresses this gap by establishing validated quantum mechanical (QM) protocols, developing a structure–reactivity framework, and delivering an open-source database to support design of PFAS degradation strategies.

First, degradation mechanisms of PFAS using chemical redox and enzymatic systems were reviewed, along with current in silico design strategies for enzymes. It was found that a protocol for design would most likely rely on metal-ion based enzymes, redox potential studies, and rational active-site engineering. A persistent gap is that most experimental studies have focused on only a handful of PFAS. In the absence of experimental data, computational studies of PFAS degradation depend heavily on DFT recommendations. However, PFAS exhibit electronically unique characteristics, and until now the field has lacked a thorough, systematic assessment of QM methods used to model them.

To address this gap, a comprehensive benchmarking study was conducted featuring 47 methods across three major basis set families. Key properties such as bond dissociation energies, ionization potentials, and electron affinities were evaluated across multiple families of PFAS and their protonation states. Benchmarking revealed method and basis set dependencies in PFAS, the reliability of property predictions, and highlighted challenges in modeling electron affinity. Overall, ωB97X-V/def2-TZVPD provided the best balance of accuracy and scalability.

Building on these validated protocols, an automated computational pipeline was developed to expand PFAS property coverage and systematically map degradation-relevant descriptors across structural classes and protonation states. This large-scale analysis identified competing bond cleavage pathways and revealed systematic, protonation-sensitive electronic distinctions across PFAS classes, head groups, and chain lengths, including distinct behavior in ultra-short-chain species. Collectively, the studies in this work establish a unified, descriptor-based framework linking molecular structure to degradability across diverse PFAS families and an open-access QM PFAS database intended to support future modeling and remediation strategies.

This talk offers an overview of the role of Monte Carlo simulations in high-energy physics and within the ATLAS experiment, highlighting its importance for detector modelling and physics analyses. It introduces the fundamental principles of Monte Carlo methods with a focus on Geant4, complemented by practical examples demonstrating its use in HEP applications, from geometry description to synthetic raw ATLAS data. Additionally, we introduce FullSimLight, a standalone framework that enables Geant4 simulations outside the full ATLAS software environment. The seminar concludes with a discussion of recent optimisation efforts in ATLAS Full Simulation for Run 3 and the High-Luminosity LHC, highlighting improvements in performance and computing efficiency.

Sahar Hosseini is an architectural and landscape historian and Assistant Professor in the Department of History of Art and Architecture at the University of Pittsburgh. Grounded in dialogues between architectural history and the environmental humanities, her scholarship positions Persianate societies at the intersection of regional and transregional mobilities, local practices, and natural contexts. She is the author of articles on Safavid architecture, and her forthcoming monograph, Zayandehrud and Its City: Reading the Riverine Landscapes of Seventeenth-Century Isfahan, presents Isfahan’s river, Zayandehrud, as central to the imperial project of refashioning the city as the Safavid Capital.

Abstract: Subtle changes to the structure of a molecule are known to substantially impact its properties. My group has initiated two research directions that involve making precise modifications to a molecule. The talk will begin with the development of atom swapping reactions that result in the substitution of a heavy atom in a cyclic compound. The second part of the talk moves into three-dimensional space with macrocyclic metacyclophanes, which are common subunits in natural products and pharmaceuticals. We will discuss how one- or two-carbon modifications lead to significant changes in rotational barriers in these non-canonical atropisomers and demonstrate how a precise understanding of what is required for atropsiomerism enables the design of new macrocycles with complex topologies.

Abstract:

In the Southern Puna Plateau, NW Argentina, Miocene to Quaternary lithospheric foundering is inferred from a combination of backarc volcanism and geophysically imaged detached lithosphere. Anomalous tectonic extension or transtension may be key to better constraining the dynamics of foundering, but the timing and scale of such extension remains enigmatic. We present new mapping and geochronology that document Middle to Late Miocene extension of the Antofalla Basin, which is centrally located in the area affected by foundering.
The Antofalla Basin is a ~125 km long, ~800 m deep linear basin that trends NNE within the Southern Puna Plateau and is internally drained. New geologic mapping, stratigraphy, and geochronology from an exceptionally well-exposed part of the Antofalla area reveal a normal fault system that became active between 13 and 11 Ma and probably generated the relief observed in the modern basin.
The onset of extension >11 Ma at the Vega Antofalla locality is consistent with age estimates from the southern part of the Antofalla Basin based on the ages of basal evaporite strata within the basin, suggesting a common evolution for the entire basin. Extension of the Antofalla Basin several Myr before major backarc volcanism (ca. 8 Ma) is consistent with numerical models of lithospheric foundering.

Individuals with less than five years of full-time service MUST be notified of non-renewal at least 3½ months prior to the end of their current contract.

Generative AI has rapidly moved from a research curiosity to a force reshaping the modern workplace. Popularized by large language models such as ChatGPT, this technological shift challenges long-standing assumptions about human labor, productivity, education, and decision-making. This presentation provides a practical overview of artificial intelligence, tracing its evolution from early statistical models and expert systems to today’s power-hungry, transformer-based generative models. It distinguishes among major AI categories—predictive AI, robotic process automation, computer vision, natural language processing, and generative AI—and examines foundational models, small language models, and task-specific systems, highlighting the absence of a single “best” model for all use cases.
Drawing on real-world applications across software development, business operations, healthcare, education, and research, the talk frames generative AI as a co-pilot that augments human creativity and productivity rather than a fully autonomous replacement. At the same time, it critically addresses risks including job displacement, bias, misinformation, privacy concerns, and over-reliance on automated systems. Emerging trends such as agentic AI, “vibe coding,” and domain-specific small models signal a shift from hype toward pragmatic deployment.
The presentation concludes by arguing that the future impact of generative AI will depend less on technological capability than on societal choices—how we retrain workers, redesign education, establish ethical frameworks, and align human agency with increasingly capable machines.

Attention has become a key normative value of the cultural present. For problems as monumental as global warming or as ordinary as the lure of transformative habits, more attention is often prescribed as a solution for impasses of agency amid the urgencies of too-late capitalism. And yet attention—a concept that unites sensory focus with time—has been undertheorized in musical aesthetics. While histories and theories of attention tend to emphasize broad epochal shifts (the accelerations of modernism, the zany incongruities of postmodernism), this talk instead considers the understanding of attentional deficiency as an inability to produce complete actions. Attention thus becomes a metric that illuminates the relations between work and our affective perception of “complete” works, which seem to convey, in their formal self-sufficiency, the dream of labor’s realization.

The talk engages Tierra Whack’s 2018 visual album Whack World, which consists of fifteen one-minute songs that Whack initially uploaded to Instagram’s video platform. Unable to muster the attentive resources necessary to complete a “finished” album, Whack produced instead what I call song stubs: songs consisting often of a single verse followed by one or two lines of the chorus, which are then sometimes truncated in mid-sentence. While several critics referred to the songs as incomplete, I argue instead that they belong to a mode of contemporary cultural production about completeness—that is, about the normative forms of attention required to produce an amount of labor that is recognizable as (a) “work.”

While medicalized discourses of attention frame its temporality as either normal or disordered, this paper explores how the musical creation of temporal experience offers a methodology for theorizing attention deficit and normativity through form in contemporary life.

Bringing together two highly regarded public intellectuals with deep experience in Indian politics and administration, this panel discussion focuses on democracy in India today and on the challenges and opportunities that will shape its future.

Mr. Amitabh Kant is a retired 1980 batch Indian Administrative Service Officer who has served as India’s G20 Sherpa from 2022 to 2026. He has also served as CEO of NITI Aayog, a public policy Government of India think tank.

Mr. Yogendra Yadav is political activist, psephologist, and a founder of the Swaraj India party. From 2004 to 2016 he was Senior Fellow at Center for the Study of Developing Societies in Delhi. He is a former member of the University Grants Commission of the Government of India.

Commentary by Mr. Kant and Mr. Yadav will be followed by questions from Professor Joseph S. Alter, Professor TK Vinod Kumar and Professor Michael Kenney as well as from members of the audience.

Public Health Genomics: Where Are We Heading?

At the end of 2024, I retired from the Centers for Disease Control and Prevention (CDC) after a 40-year career in public health. For the past 26 years, I led the Office of Public Health Genomics (PHG), which was formed in 1998. During my career, I was lucky to witness and contribute to the development of three fields: genetic epidemiology, public health genomics, and precision public health. Together, these fields are increasingly crucial in realizing the population health benefits of genome discoveries and other new technologies. But we are still in the early days on the trajectory from genes to public health. With many global challenges ahead, the next 40 years are going to be more exciting than the last 40 years in applying these fields to emerging validated tests and interventions that can save lives, prevent disease, and reduce health inequities. Many of the advances in genomics translation and implementation could be unravelled because of the current turmoil and budget cuts in science and public health in the United States. In this talk, I offer a brief perspective on the evolution and contributions of these fields to medicine and public health and offer some recommendations to increase their population health impact.

Jonah Belback Presentation Information

Title: Large Language Models and Domain Knowledge for Enhanced Multimodal Learning in Predicting Breast Cancer Recurrence Risk

Abstract: Despite the effectiveness of adjuvant endocrine therapies such as Tamoxifen and Aromatase Inhibitors, their long-term use and association with significant side effects causes poor adherence for patients unsure on their individual benefit. In order to address this issue compromising patient recovery, an early response biomarker/method measuring the clinical efficacy of Tamoxifen and Aromatase is needed. In this
work, we proposed a novel method of autonomously integrating domain-specific knowledge into a Transformer-based model to facilitate multimodal learning by leveraging large language models (LLMs) and medical domain knowledge represented by textual data through semantic embeddings and knowledge graphs for enhanced recurrence risk prediction in estrogen positive breast cancer patients. We evaluated the performance of our proposed method on a dataset of 679 patients. Our proposed models achieved superior AUC scores when compared to baseline models, in distinguishing recurrence-free patients from those with new primary cancers. This study showed the promise of our novel technical methods and proved the concept that LLMs and domain knowledge can be a useful source of intelligence to enhance AI models, which is a significant advance for multimodal AI learning. With our enhanced prediction models, it can minimize unnecessary harms from Tamoxifen or Aromatase treatments for patients unlikely to benefit from them and increase patients’ adherence for those more likely to benefit.

Title: Script Gap: Evaluating LLM Triage on Indian Languages in Native vs Roman Scripts in a Real-World Setting

Abstract: Large Language Models (LLMs) are increasingly deployed in high-stakes clinical applications in India. In many such settings, speakers of Indian languages frequently communicate using romanized text rather than native scripts, yet existing research rarely evaluates this orthographic variation using real-world data. We investigate how romanization impacts the reliability of LLMs in a critical domain: maternal and newborn healthcare triage. We benchmark leading LLMs on a real-world dataset of user-generated queries spanning five Indian languages and Nepali. Our results reveal consistent degradation in performance for romanized messages, with F1 scores trailing those of native scripts by 5-12 points. At our partner maternal health organization in India, this gap could cause nearly 2 million excess errors in triage. Crucially, this performance gap by scripts is not due to a failure in clinical reasoning. We demonstrate that LLMs often correctly infer the semantic intent of romanized queries. Nevertheless, their final classification outputs remain brittle in the presence of orthographic noise in romanized inputs. Our findings highlight a critical safety blind spot in LLM-based health systems: models that appear to understand romanized input may still fail to act on it reliably.

Bio: Manurag is a first-year PhD student in the Intelligent Systems Program. His research interests include multilingual NLP, responsible AI, and AI for social good. Previously, he was a Data Science for Good Fellow at the University of Washington, Seattle Campus, and worked as a technology policy analyst with NITI Aayog, the Government of India’s apex think tank. He holds a master’s degree from the University of Pennsylvania and completed a thesis on explainable AI under Professor Li Shen.

Title: “No only to anti-realism”: Some skeptical thoughts on scientific realism

Abstract:

Debates about scientific realism and anti-realism have been a prominent part of the landscape of philosophy of science for the past few decades, which have seen a proliferation scientific realisms and anti-realisms. Nearly 40 years ago Howard Stein, from whom I borrow my title, added a skeptical voice to these discussions, arguing that the issue between scientific realists and its opponents had not been clearly drawn. This talk takes up that skeptical thread. I will argue, first, for the modest conclusion that we should believe whatever we have sufficiently good evidence for. As this includes the existence of some things (including atoms) that are not directly observable, this modest conclusion involves rejection of any form of anti-realism that involves a prohibition against accepting the existence of unobservable entities. A mere absence of a prohibition, however, hardly deserves to be elevated into a philosophical position, hence I don’t consider my view to be adding to the menagerie of versions of “scientific realism.” I doubt that there is any defensible position worthy of that name.

Adolf Grünbaum Memorial Lecture

Adolf Grünbaum was the first Andrew Mellon Professor of Philosophy at the University of Pittsburgh from 1960, when he was hired away from Lehigh, until his death in November 2018. He was the world preeminent philosopher of physics of his generation, and he was instrumental in building philosophy and philosophy of science at Pitt. Among other things, Adolf founded the Center for Philosophy of Science in 1960, and remained its director until 1978. Adolf also inaugurated the Annual Lecture Series in 1961, and so this is the 66th occurrence of this lecture series.

His works include very important books such as Philosophical Problems of Space and Time (1963) and The Foundations of Psychoanalysis (1984). In honor and memory of his legacy of prolific and profound contributions to the field of philosophy, the Center has established an annual Adolf Grünbaum Memorial Lecture thanks to a gift from his daughter Barbara Grünbaum and her family. We are extremely grateful for this gift. We are delighted that this year the Adolf Grünbaum memorial lecture will be given by Professor Wayne Myrvold. Dr. Deborah Studen-Pavlovich, DMD, served as professor, chair, and residency program director of the Department of Pediatric Denistry at the University of Pittsburgh School of Dental Medicine. In this role, she oversaw all of the Department's curricula and activities, including the annual Give Kids A Smile Days. She graduated cum laude from Butler University in 1976 and received her DMD and certificate in pediatric denistry at the University of Pittsburgh School of Dental Medicine.

Dr. Alema Galijatovic Idrizbegovic, PhD, was born in Sarajevo, Bosnia and Herzegovina. In 1991, she came to the United States as a high school exchange student in Gallatin, TN. After obtaining a postdoctoral fellowship at the University of California, she started her career at Merck & Co. in 2003, where she held several positions and reached her most recent post as the Vice President of the Nonclinical Drug Safety Department.

Affirmation & Erasure: Lessons from the Architecture of the Irish Room
Presented by Lovemerry Illin, a December 2025 graduate of the University of Pittsburgh with a Bachelor of Arts in Architectural Studies and a minor in Studio Arts. A recipient of the Mary Campbell Cross scholarship, Lovemerry conducted extensive archival research on the Irish Nationality Room, which funded a two-month unpaid internship at a preservation architecture firm in Dublin, Ireland. Her commitment to the field was recently recognized by the Young Preservationists Association (YPA) of Pittsburgh, where she was honored with the Dan Holland Promise Award for exemplary early-career work in Historic Preservation. Lovemerry currently interns with the YPA, managing finances and event programming, and plans to pursue a Master’s degree in Historic Preservation to continue her professional development in the stewardship of the built environment.

Come join us for coffee and an opportunity to get acquainted with Karen Sebolt, Director of Heinz Memorial Chapel. Sam Young, Co-Vice Chair of Staff Life, will engage in a conversation with her to discuss her life and career at Pitt!

Karen Sebolt earned an MBA from the University of Pittsburgh School of Business, subsequently gaining a CPA license and working for three large health systems in audit, finance and performance improvement during the 1990’s hospital mergers.

Three small children, dual-career travel demands, and 2400 miles from family support, she made the decision to step away from 15 years in the professional world. The next 15 years she volunteered in the public schools, local church, art & music programs, and Girl Scouts; while directing family relocations from Texas to California to Virginia and finally back to Pittsburgh.

In 2017, ready to thoughtfully reenter the workforce, Karen accepted a part-time docent position at Heinz Chapel. During the early days of the pandemic, she was asked to lead Heinz Chapel, write public health and safety measures, and reopen the Chapel for wedding couples.

Today, Heinz Chapel is a natural fit with the Hospitality division within Business, Hospitality and Auxiliary Services (BHAS). Under Karen’s leadership, the Chapel team focuses on innovative programming to engage students and the broader community, operational efficiencies to support the mission and stewardship of an architectural landmark, and continued excellence in customer service.

The opportunity to return to her alma mater and work in one of its most treasured buildings represents a meaningful full-circle achievement. Her highlight of the year is presenting the Women in the Windows tour in March—an experience she developed to highlight the remarkable women represented in the Chapel’s stained-glass and to celebrate their lasting impact.

Abstract:

Dimension reduction methods extract meaningful representations from high-dimensional data but often operate unsupervised, ignoring outcome information that is crucial for prediction and inference. This dissertation develops supervised extensions of widely used methods, including nonnegative matrix factorization (NMF) and multidimensional scaling (MDS), to enhance interpretability, mitigate confounding, and improve predictive performance in precision medicine involving large biomedical, clinical, and omic data sets.

The first part of this dissertation proposes an outcome-driven supervised NMF method for estimating individualized treatment rules by integrating structured data and unstructured text. Unlike conventional NMF, our approach factorizes document representations while directly optimizing treatment decisions. Through simulations and application on MIMIC-IV intensive care unit data with radiology notes, it identifies predictive but less prevalent topics, refines broad topics, and improves classification accuracy over standard approaches.

The second part of this dissertation focuses on supervised dimensionality reduction for microbiome data. Principal coordinate analysis (PCoA) is a common approach that provides low-dimensional representation of the data but does not incorporate outcome information. We propose a supervised and covariate adjusted PCoA algorithm that integrates outcome information while reducing bias from demographic, lifestyle, and technical variations. Our method produces low-dimensional representations that better separate disease outcomes in visualizations and improve predictive accuracy. We demonstrate its effectiveness through simulations and applications to several metagenomic profiling studies, highlighting its ability to extract meaningful microbiome-outcome associations while mitigating confounding effects.

The third part of this dissertation extends supervised NMF for the integration of data from multiple microbiome studies. Despite the promise of large-scale microbiome studies for identifying robust disease signatures, small sample sizes and study-specific batch effects pose substantial analytical challenges that existing integration methods do not fully address. Moreover, existing methods emphasize unsupervised integration. Our approach models shared microbial structures across studies through a common latent factor space, while estimating study-specific parameters to account for batch-induced variation, and jointly predicts health outcomes. Through simulations and application to microbiome profiling studies, we demonstrate our method's ability to recover generalizable microbial signatures and improve outcome prediction across heterogeneous studies.

Public health significance: This dissertation develops supervised dimension reduction methods to improve
prediction and interpretation in high-dimensional biomedical data. By integrating outcome information into analyses of clinical text and microbiome data, the proposed methods support more accurate individualized treatment decisions and disease risk estimation, thereby advancing biomedical and public health research.

All cells sense and respond to physical and chemical cues in their environments. They accomplish this through signal transduction systems—networks of interacting proteins that detect and interpret specific input signals and control appropriate cellular responses. In bacteria, these systems are found in remarkable numbers within individual organisms and across different bacterial species. They play a central role in regulating basic aspects of microbial physiology and mediate responses to diverse environmental signals. I will describe work in which we have explored the organization and properties of these circuits in the well-studied and genetically tractable organism Escherichia coli.

Federal research funding is often treated as neutral and meritocratic, yet it powerfully shapes who gets to produce educational knowledge and whose expertise counts. In this talk, Dr. McCambly will present results from her analysis of education research grants from IES and NSF between 2008 and 2022. She will show that Minority-Serving Institutions are persistently underfunded in standard research competitions, even when they look similar to white-serving institutions on capacity and research mission. The only policies that shift this pattern are explicitly race-conscious, institution-targeted capacity-building grants—policies that are symbolically positioned outside “real” research. The talk argues that by framing redistribution through institutional capacity deficits rather than expertise, these designs achieved equity gains while simultaneously rendering themselves legible as non-meritocratic—and therefore politically expendable in the current ideological climate. The tensions surfaced by these findings will be used to guide dialogue with participants in the space.

Title: Hot to the Touch and Chilled to the Bone

Abstract:

In 1896, the Swedish physiologist Torsten Thunberg reported a curious discovery: when he touched a grid of alternating warm and cold brass pipes, the sensations of warmth and cold on his hand somehow ‘fused together’ into a ‘special sensation of heat’ — a sensation that many experimental subjects since then have described as painful. Since the 1990s, this so-called ‘thermal grill illusion’ (TGI) has become the conventional method of investigating ‘illusory pain’, and is said to hold important insights for our understanding of the bodily senses and clinical pain pathologies. However, what exactly these insights are meant to be remains largely unclear: indeed, after more than a century of scientific interest in Thunberg’s discovery, the sensory fusion he described is about as puzzling as ever. In this talk, I aim to articulate why the thermal grill illusion is a puzzle worth caring about, why it has been so difficult to solve, and what, if anything, we can learn from putting our hand on a grill.

While many students are habituated to think of slavery as a “Southern” institution and are taught that Pennsylvania “abolished” slavery in 1780, the reality is that in Western Pennsylvania over 2700 unique heads of households were enslavers between 1780-1850. These enslavers were often the most powerful people in society: generals, politicians, judges, sheriffs, landowners, and business owners. They were also ministers, elders, and trustees of prominent institutions. This expansive and ongoing research project connects these enslavers with the religious institutions that materially benefited from their stolen wealth (churches and universities) and maps how they continue to be memorialized in Pittsburgh’s built environment (e.g, street names, monuments, neighborhoods, etc.). It will further reflect on how an epistemology of ignorance, or intentional forgetting, of those who were enslaved functions within our cultural and institutional memory. For the purposes of this colloquium, special focus will be placed on enslavers who had ties to the University of Pittsburgh.

Jill Joshowitz is a scholar of Jewish visual culture in the late antique Eastern Mediterranean and the author of Visual Exemplars: The Art of Biblical Figures in Jewish Art and Literature in Late Antiquity (Brill, 2025). She has curated exhibitions for Jewish cultural institutions throughout the United States including the Maltz Museum, Museum of Jewish Heritage, and the Yeshiva University Museum.

Abstract:

The rapid expansion of multi-omics and single-cell technologies has opened new opportunities to unravel complex biology and advance precision medicine. However, fully leveraging these technological advancements requires innovative statistical and computational methodologies capable of handling the inherent complexity of omics data, including high dimensionality, strong correlations, and sample heterogeneity. This dissertation develops and benchmarks novel statistical approaches designed specifically for multi-omics mediation analysis and single-cell copy number variation (CNV) inference.

In the first part, I developed a statistical framework for high-dimensional mediation analysis under the assumption of homogeneous mediation effects across patients. Current mediation methods face significant challenges, including reduced statistical power due to unknown signal structure and difficulties in prioritizing correlated mediators. To address these challenges, I proposed the Partial Sum Statistic with Sample Splitting Strategy (PS5) method. PS5 employs a novel partial sum statistic that enhances statistical power for detecting global indirect effects under varying mediator signal strengths. Additionally, a multiple sample splitting strategy robustly selects correlated mediators, ensuring stable identification even in highly correlated omics data. Extensive simulations and application to the COPDGene cohort demonstrate its ability to identify lung imaging biomarkers mediating genetic and environmental
effects on lung function, providing clinically meaningful insights for COPD.

In the second part, I addressed the urgent need for large-scale reliable benchmarking of scRNA-seq CNV inference tools. While numerous scRNA-based CNV inference tools have been developed, conclusive guidance for tool selection remains incomplete. I designed a benchmarking study that comprehensively evaluates twelve scRNA-based CNV inference tools across 28 real datasets and a series of synthetic datasets, including primary tumors, metastases, cell lines, and patient-derived organoids. Our extensive benchmarking assesses critical factors such as malignant cell classification accuracy, CNV inference precision, computational scalability, tool usability, and parameter robustness.

The results show that method performance depends strongly on data type and allelic signal quality: Numbat performs best when allelic signal is strong, while expression-only tools such as CopyKAT performed reliably in settings without raw sequencing data. Based on these results, we provide practical, scenario-based guidelines to help users select appropriate tools and parameters.

The third part of this dissertation focuses on the development of a comprehensive software package for p-value combination. P-value combination methods aggregate evidence across multiple related tests, improving statistical power, and enabling valid global inference in high-dimensional and multi-omics settings. Although many combination methods have been proposed, user-friendly and unified software implementations remain limited. In this study, we implement 29 combination methods, which can be grouped into three categories: (1) combining independent p-values, (2) combining dependent p-values, and (3) conjunction and partial conjunction tests.

While the first two categories focus on detecting at least one true signal, conjunction-based methods assess whether all or a specified subset of hypotheses are non-null. Simulation results show that certain methods, such as Edgington’s method, arithmetic averaging, and geometric averaging, can easily under- or over-control type I error.

Public health significance: Collectively, these methodologies enhance the integration, interpretation, and clinical relevance of multi-omics and single-cell data. By identifying actionable biomarkers, improving data reliability, and revealing distinct patient subgroups, our research advances precision medicine, facilitates targeted therapeutic strategies, and deepens understanding of molecular heterogeneity in complex diseases.

About the Presenter:

Jesko Hirschfeld studied economics at the universities of Bonn, Frankfurt am Main, and the Free University of Berlin. From 1996 to 1998, he worked as a scientific staff member for the German Federal Parliament (Bundestag), and from 1998 to 2002, he worked on his doctorate with a scholarship by the German Research Foundation (DFG) in the Research Training Group “Agriculture and Environment” at the University of Göttingen. Since 2002, he has been a research associate and later subject area manager for water and land management at the Institute for Ecological Economy Research (IÖW) in Berlin in the research field “Environmental Economics and Policy.” From 2017 to 2020, in addition to his work at the IÖW, he was a visiting professor for landscape economics at the Technical University of Berlin. From 2022 to 2024, he taught as a visiting professor of environmental economics at Humboldt University in Berlin. He has worked on numerous inter- and transdisciplinary projects on natural climate protection, nature-based adaptation to climate change, land and water use, and integrated coastal zone management, focusing primarily on economic aspects and ecological-economic assessments. In recent years, he focused on urban ecosystem services provided by urban green, parks, and gardens.

During the session, Dr. Liz Arkush (Anthropology) will present a book chapter-in-progress, tentatively titled “Those who most pursued their freedom”: Archaeological and ethnohistoric perspectives on Colla resistance to Inca rule. The chapter examines Colla resistance to Inca expansion in the Lake Titicaca region by comparing written sources with archaeological evidence, highlighting both points of convergence and disagreement. Together, these sources reveal a prolonged and violently contested process of Inca expansion, marked by fortified resistance, eventual abandonment of hillforts, and the construction of Inca structures that may have commemorated final conquest.

Abstract:

Vast regions of Arctic tundra are characterized by ice wedge polygons, geometric features created by the repeated freezing and thawing of the ground surface. Troughs form between polygons, ranging from half a meter to three or more meters across. These troughs create networks of microtopographic drainages, with varying degrees of complexity and connectivity, as water pools or runs off across the landscape. These "capillary" networks are the product of complex interactions between hydrology, permafrost thaw, and vegetation, with the potential to either accelerate degradation permafrost and transport of material off the landscape, or, in some cases, stabilize underlying permafrost and reduce surface runoff. Ultimately, the degree of connectivity on this microtopographic scale may alter not only localized processes, but the delivery of water and organic matter to streams and rivers across the Alaskan North Slope.

This two-day symposium will convene scholars of graffiti and street art in Latin America, South America, East Asia, and the Middle East to offer a fuller telling of graffiti’s past and present.

The program includes a series of roundtables where presenters will offer short works in progress and be in dialogue with colleagues at the University of Pittsburgh. These discussions will be framed by a “gallery” of photographs of graffiti and street works taken by participants from all over the world, courtesy of the World History Center.

Additional events include a hybrid student-meets-scholar session pairing graduate students with mentors to share works in progress, as well as a short book launch for the co-authored volume Creative Cities, Tourism and Street Art in a Global Frame: The City as Discovery. Attendees are also invited on a walking tour of local street art and graffiti in Pittsburgh’s Bloomfield and Garfield neighborhoods, co-led by Professor Bruce and Hemispheric Conversations Urban Art Project.

Abstract: The development of an atomistic understanding of heterogeneous catalysis has transformed materials discovery: from the Edisonian approach that led to the Haber-Bosch process, to the modern physics-informed framework that allows prediction of the optimal catalyst composition before an experiment is performed. Extending this framework to interfacial charge transfer processes—such as corrosion, energy conversion and storage, electrochemical fuel synthesis—remains challenging. Experimentally, probing the electrified double layer with atomic resolution is extremely difficult, while simulations are limited by the self-interaction error (SIE) and its associated problems intrinsic to semi-local density functional theory (DFT). Most relevant to this talk is the unphysical delocalization of electron density, which results in unacceptable errors when describing ions at this level of theory.

In this talk, I will share our group's efforts to overcome these challenges and extend DFT's ability to capture both charge transfer thermodynamics and kinetics. First, I will introduce a Born-Haber cycle approach for modeling corrosion thermodynamics that avoids explicit treatment of corrosion products. Using this framework, we investigated the empirically observed activity-stability tradeoff in water electrolyzer anodes and found no fundamental intrinsic coupling between these two critical parameters. I will then turn to kinetics, which requires explicit treatment of ions, and will describe our efforts to establish an experimental benchmark for evaluating density functionals in charge transfer processes. I will discuss how this benchmark informs our understanding of pH effects in hydrogen evolution electrocatalysis, and its broader implications for modeling interfacial electrochemical processes.

Bio: Joe Gauthier is an Assistant Professor of Chemical Engineering at Texas Tech University. Dr. Gauthier graduated summa cum laude with his B.S. degree in Chemical Engineering at The Ohio State University in 2015 before moving to California to pursue doctoral research at Stanford University. After one year as a visiting student in the Physics Department at the Technical University of Denmark, he completed his PhD at Stanford in 2020. Before starting at TTU, he was a Postdoctoral Researcher at UC Berkeley and Lawrence Berkeley National Laboratory. He is the author of more than 40 peer-reviewed publications and leads a 12-person research team at TTU comprising a mix of Ph.D., M.S., and undergraduate students. At TTU, his research program focuses on heterogeneous (electro)catalysis, ultimately with the aim of solving problems related to renewable energy conversion/storage and decarbonization of the chemical industry.

Title: Introducing A Wisdom Framework for Science in the 21st-century: The Idea of a Two-Track Scientific Community

Abstract:

In this talk, my task will be to show how wisdom, which is an integrated form of multi-level skills, could be incorporated into the research/ teaching practices of contemporary science with some good effects. Additionally, such wisdom can allow us to use our intellectual breakthroughs to serve our existential ends. Our science today is built on the idea of developing expertise in multiple narrow domains, which then would be scaled up to yield quick flows of innovations. While a science based on this kind of narrow expertise can bring us rapid flows of novel things, it can also lead us to a crisis of sustainability, commodification of research, and a slide towards inequality. To improve matters, I suggest that our narrow practices be scaffolded by a wider and more expansive practice that could function as scaffolds to our current sets of expertise. This combination of a faster and a slower track will allow us to address the blind spots that often lie embedded in our expertise and later gives rise to evidence-in-use problems. Setting up a link between these two tracks may be called wisdom, which we badly need today to engage with our current exponential technologies and their effects on us. This of course needs the inclusion of some new members into the peer community.

Adding a wisdom track in this way to our system can help us develop new sets of research questions, training procedures, or research protocols. It can thus allow us to explore our areas of ignorance. While there is much discussion about the Freudian kind of ignorance that makes us blind to various things, there is also a Socratic form of ignorance that can be used as a springboard for new ways of thinking. In true Socratic spirit then, I claim that this wisdom approach might help us in three specific areas – in gatekeeping decisions, building trust in science, and in setting up consiliences with other knowledge communities and their traditions.

Bio: Yiqun Xie is an Assistant Professor in the Center for Geospatial Information Science, Dept. of Geographical Sciences, and an Affiliate Faculty at the AI Interdisciplinary Institute at Maryland (AIM), at the University of Maryland. He received his PhD in Computer Science at the University of Minnesota, and his research addresses fundamental challenges facing AI for spatio-temporal data and related scientific problems. His current work focuses on advancing use-inspired AI models with explicit geo-awareness and knowledge-guidance to improve model generalizability over space and time for large-scale applications. He is leading multiple interdisciplinary AI projects funded by NSF, NASA, and Google, with research published in both top AI and domain science venues. His work has been recognized by multiple best paper awards from IEEE ICDM 2021, ACM SIGSPATIAL 2025 and 2019, SIAM Data Mining 2023, SSTD 2019, and was highlighted by the Great Innovative Ideas program by CCC at CRA. He delivered invited panel talks on GeoAI for committee meetings of the National Academies on Science, Engineering, and Medicine (NASEM) and the National Geospatial Advisory Committee (NGAC).

Abstract: Crystallization plays distinct, high‑value roles across the drug discovery and development continuum - from a decisive compass for structure‑guided medicinal chemistry to an enabling workhorse of process chemistry. This dual utility will be illustrated through two case studies. First, an industrial synthesis of an antimalarial clinical candidate showcases a fully crystallization‑based route eliminating all chromatographic purifications. Strategic discovery of multiple crystalline intermediates spanning a diastereoselective Claisen rearrangement/salt resolution, stereoselective iodolactonization, and stepwise formation of a densely substituted diazabicyclo[6.2.0]decane system enabled clinical entry of an antimalarial with a novel mechanism-of-action. Second, in a medicinal chemistry setting targeting an immune receptor, iterative X‑ray co‑crystallography transformed a historically shallow, “undruggable” sialic acid binding site into a ligandable pocket and led to discovery of ligands with nanomolar affinity.

ABSTRACT:

Genetic variation underlies much of human diversity, including differences in disease severity, progression, and response to therapy. Yet most mouse models used to study genetic diseases and evaluate therapeutic interventions are maintained on a single genetic background, limiting their ability to reflect the genetic heterogeneity observed in patients. This limitation extends even to Mendelian disorders such as LAMA2-deficient congenital muscular dystrophy (LAMA2-CMD), where individuals with the same pathogenic mutation in the LAMA2 gene often exhibit variable clinical presentations. Current preclinical mouse models of LAMA2-CMD, constrained by their genetic uniformity, fail to capture this variability.

To address this, we generated genetically diverse LAMA2-CMD mouse models using the BXD recombinant inbred panel. The Lama2 c.271+1G>A mutation, traditionally maintained on a C57BL/6J background, was introduced onto three additional genetic backgrounds: DBA/2J (CMD-D2), BXD-67 (CMD-67) and BXD-87 (CMD-87). We assessed body weight, forelimb grip strength, locomotor activity, muscle contractile force, histology and transcriptomics across all strains.

Failure to thrive and impaired muscle contractile force were consistent across strains, indicating strong mutation-driven effects. In contrast, grip strength, locomotor activity, and muscle fiber pathology showed strain-dependent differences that became more pronounced at advanced disease stages. CMD-67 exhibited an early muscle fiber size reduction at 8 weeks followed by convergence toward wildtype distributions by 25 weeks, unlike the more progressive atrophy observed in other strains. Transcriptomic profiling revealed a shared immune-dominant signature across all strains at 8 weeks, with the CMD-67 mice showing concurrent enrichment of muscle-associated pathways. By 25 weeks, transcriptional divergence became more apparent: CMD-B6 and CMD-67 demonstrated co-activation of immune and muscle regenerative programs, CMD-D2 exhibited a predominantly immune-activated profile, and CMD-87 showed enrichment in muscle remodeling pathways. In contrast, downregulated pathways were highly strain-specific at both time points.

Overall, while core dystrophic features are largely mutation-driven, disease severity, progression, and molecular signatures are strongly modified by genetic background. This work establishes a novel LAMA2-CMD mouse panel as a valuable resource for future studies, including identification of disease modifiers and evaluation of therapeutic responses, and supports the premise that incorporating genetic diversity into preclinical models may improve translational relevance to human disease.

This year, all of the proceeds raised from our show will be donated to the Pittsburgh Foundation and the Cauliflower Alley Club. From their website, the Pittsburgh Foundation works to improve the quality of life in the Pittsburgh region by evaluating and addressing community issues, promoting responsible philanthropy and connecting donors to the critical needs of the community. The Cauliflower Alley Club raises funds for veteran wrestlers struggling financially with items like medical expenses, funeral arrangements, and other needs.

Abstract:

In clinical trials, response-adaptive randomization (RAR), or outcome-adaptive randomization, aims to achieve greater patient benefit by assigning more patients to more efficacious treatments, while maintaining the validity of treatment comparisons. In contrast to RAR, which focuses on improving patient outcomes by adjusting treatment allocations, a dynamic treatment regime (DTR) offers a structured approach to decision-making over multiple stages. Sequential multiple assignment randomized trials (SMARTs) provide a useful framework for constructing DTRs and providing unbiased between-DTR comparisons. A limitation of SMARTs is that they do not utilize data from past patients, which could help reduce the likelihood of exposing new patients to inferior treatments. In practice, this may lead to reduced treatment adherence or patient dropouts. Moreover, identifying the optimal DTR with censored survival data remains a significant challenge. In this dissertation, we compare the performance of various estimators for estimating causal treatment effects using different estimands in RAR designs, introduce the outcome-adaptive randomization in the SMART design, and propose a new statistical method for estimating the optimal DTR with censored survival data.
In the first part of the dissertation, we propose estimators of causal treatment effect for RAR designs using inverse-probability-weighting (IPW) and compare them analytically to the sample average treatment effect (ATE) on bias, variance, type I error, and statistical power. Extensive simulation studies were conducted to assess the performance of these estimators. The results show that, when implemented and analyzed correctly, RAR can treat a substantial proportion of patients using the superior treatment while minimally sacrificing power compared to standard allocation designs. Both analytical and simulation results indicate that the IPW-based estimators are consistent, have a smaller bias, and are more efficient than the sample ATE. Moreover, the methods based on log relative risk and log odds ratio better control the type I error than the sample ATE-based method.
In the second part of the dissertation, we propose a generalized outcome-adaptive (GO) SMART design that adaptively skews stage-specific randomization probabilities toward treatments observed to be more effective in previous patients. To correct for bias induced by outcome adaptive randomization, we develop G-estimators and inverse-probability-weighted estimators of DTR effects embedded in a GO-SMART and show analytically that they are consistent. Simulation results show that, compared to a traditional SMART, RA-SMART (Wang et al., 2022), and SMART with adaptive randomization (Cheung et al., 2015), a GO-SMART design treats significantly more patients with the optimal DTR and achieves higher overall response rates while maintaining similar or better statistical power.
In the third part of the dissertation, we propose a tree-assisted imputation-based Q-learning (TAI Q-learning). Q-learning is a commonly used and straightforward reinforcement learning algorithm for determining the optimal DTRs with right-censored survival data. However, it is sensitive to model misspecification, which is a known limitation. In TAI Q-learning, double robust survival trees and tree ensembles are used to estimate the optimal treatment rules at each stage, and Multivariate Imputation by Chained Equations (MICE) is used to predict the optimal survival time. Through extensive simulation studies, we compare the performance of traditional Cox proportional hazards (Cox PH) and Accelerated failure-time (AFT) models with the non-parametric tree-based methods during the optimization step, and we compare Hot-deck multiple imputation with MICE in the imputation step. The simulation results show that MICE is easier to implement and often outperforms Hot-deck imputation. Moreover, in multilevel treatment scenarios, tree-based methods outperform Cox PH and AFT models in estimating optimal treatment rules.
Public health significance: The design and statistical methods proposed in this dissertation will increase the acceptability of adaptive treatment assignments, thereby reducing the rates of noncompliance and dropout. Additionally, the contributions to the field of optimization in sequential decision making have a substantial impact on identifying optimal therapies for chronic diseases such as AIDs, mental illnesses, substance abuse, and many cancers. Consequently, this benefits patients enrolled in trials and ultimately improves public health and safety.
