Dr. Martina Anto-Ocrah is an Assistant Professor of Medicine & Epidemiology whose research centers on pregnancy, sexual, and reproductive health epidemiology in the United States (US) and global populations. As a reproductive epidemiologist, Dr. Anto-Ocrah’s US-based work explores the impact of traumatic brain injuries on women’s reproductive wellbeing (pregnancy, sexuality, menstruation, socio-cultural norms, etc.), while her global health work is focused on obstetric emergencies and The Three Delays. Though a quantitative methodologist, Dr. Anto-Ocrah has a deep appreciation for mixed methods research and often involves qualitative research methodologies and machine learning approaches in her work.

Abstract: One research strand in this thesis focuses on advanced student sensemaking in upper-level electrostatics, while solving problems involving the electrostatic potential using Laplace’s equation or the method of images. Individual interviews were conducted and student sensemaking was analyzed using the epistemic game framework proposed by Tuminaro and Redish. Findings highlight the nested epistemic games played by students and shed light on the sensemaking of students while solving problems in advanced contexts.

To support students in solving electrostatic boundary value problems, guided inquiry-based tutorials were developed and validated for problems that can be solved appropriately and effectively using Laplace’s equation or the method of images. These tutorials were implemented across three instructors’ classes, along with unscaffolded and scaffolded pre and posttest. Think-aloud interviews with advanced students were also conducted. Although students showed gains in the scaffolded tests, greater learning gains were observed on unscaffolded versions of tests for Laplace’s equation. Findings also suggest that how instructors frame the instructional value of the tutorials may have impacted student engagement and their performance on associated assessments.

Another investigation addresses the widespread misinformation surrounding recent advances in quantum information science and technology (QIST). Interviews with college-level educators who are also quantum researchers identified challenges in QIST that lead to misinformation and strategies for helping students, especially early learners at pre-college and college levels, develop good understanding of QIST concepts so that they can avoid being misinformed.

Perception of recognition by others and identity in physics can be shaped by various forms of feedback and experiences. We discuss how course grades are related to changes in identity and perceived recognition from the beginning to the end of the course across three years for students enrolled in a calculus-based physics 1 course. Regression models predicted pre survey and post survey means and final course grades, including effects of gender and interactions between grade and gender. All student groups, on average, showed declines in identity and perceived recognition after receiving lower grades. However, women exhibited larger declines compared to men for lower grades.

Abstract: Fault-tolerant quantum computation demands many physical qubits per logical qubit, making scalability a central challenge. Topological quantum computation instead builds fault tolerance into the hardware using exotic quasiparticles such as Majorana zero modes (MZMs) that are topologically protected from local decoherence. A leading platform for MZMs is the quasi-1D hybrid superconductor-semiconductor nanowire. Realizing MZMs is fundamentally a materials engineering challenge, motivating a detailed understanding of the microscopic properties of these systems.

The experimental project in the thesis explores Sn-InAs nanowire Josephson junctions by studying Andreev bound states (ABS), which serve as direct probes of the junction’s defining properties. Sn is a strong alternative to conventional Al superconductor for its larger, hard-induced gap, expected to provide better protection against quasiparticle poisoning. This thesis presents, to our knowledge, the first ABS microwave spectroscopy measurements of Sn-based junctions.

Using circuit quantum electrodynamics (cQED) toolkit, we perform microwave spectroscopy on the junction embedded in an RF-SQUID and coupled to a coplanar-waveguide resonator. Two-tone spectroscopy maps the ABS dispersion versus flux and gate voltage, from which we estimate the mode transparency, induced gap, number of ABS modes, and their gate tunability. We resolve a unity-transparency mode that disperses towards zero energy at half flux, a trivial state resembling an MZM. We also observe signatures of spin-orbit splitting and multiple interacting ABS manifolds. Using single-tone spectroscopy, we analyze the junction’s current-phase-relation (CPR), skewness, harmonic content and critical current, and compare them with the Beenakker short-junction model.

Beyond the experiment, two theoretical proposals extend the platform from characterization to engineering functional devices. First, a single skewed ϕ0-junction can act as the nonlinear element of a three-wave-mixing parametric amplifier, providing an alternative to multi-junction SNAIL devices, using an applied magnetic field, or an on-chip micromagnet at zero field, to tune to the Kerr-free sweet spots dominated by the third-order nonlinearity. The second is motivated by the Sachdev-Ye-Kitaev model, where many MZMs interact via random, all-to-all four-body couplings. We propose an experimental realization of its smallest building block, a four-body MZM interaction, in a hybrid nanowire, and show how to detect and enhance this interaction through transport studies.

Abstract: This dissertation investigates how collaboration, reflection and inclusive practices may enhance conceptual understanding, identity development and equity in physics education. The first line of research examines how peer collaboration supports conceptual learning in graduate-level physics. Using Conceptual Survey of Electricity and Magnetism (CSEM) and Magnetism Conceptual Survey (MCS), I compared individual and group performances to understand the mechanisms of learning through discussion. Peer interaction enhanced conceptual learning through construction, in which one student knows the correct answer and guides a peer and co-construction where no group member initially knows the answer but the group collectively reasons to the correct solution. Construction accounted for most improvements, while co-construction may have promoted deeper reasoning, showing that peer discussion alone can drive meaningful learning without instructor facilitation. Subsequent surveys and interviews examined how peer collaboration shapes the conceptual and pedagogical development of graduate teaching assistants. Extending this work to undergraduate physics courses, I examined data from both algebra and calculus-based classes before and during COVID-19 pandemic. Students who collaborated, especially online, achieved higher grades and reported stronger peer influence on self-efficacy compared to those who worked alone, while prior preparation and self-efficacy continued to be strong predictors of academic performance. A third line of research explores how physics students conceptualize thinking like a physicist. Interviews with graduate students suggests that while core courses emphasized mathematical rigor, elective courses and research experiences may have more effectively supported conceptual understanding and authentic physicist identity development. Through departmental surveys and interviews, I investigated perceptions of conceptual and quantitative problem-solving. We found both alignments and discrepancies in how courses emphasize these aspects, offering insights to inform teaching and assessment practices that foster expert-like problem-solving skills. The final line of research advances inclusion in quantum information science and technology (QIST) education. Interviews with QIST educators highlighted how the field’s interdisciplinary nature can promote collaboration and diversity. Building on these insights, the QuanTime initiative introduced accessible K–12 quantum activities to improve teacher confidence and student engagement. Together, these studies demonstrate how collaborative and inclusive practices can transform physics education in general and quantum education in particular.

Abstract: This dissertation aims to address some remaining questions that cannot be explained solely with the Standard Model of particle physics. The investigations proceed along two fronts: 1) the development of novel phenomenological frameworks for new physics searches at energy and intensity frontiers that will enhance and broaden the impact of experimental analyses, and 2) the construction of experimentally testable models to explain puzzles like origin of the neutrino mass and the electroweak hierarchy problem. In the first direction, in order to optimize new physics searches at experiments, we study and formulate simplified frameworks in which the new physics signatures are characterized by variables most directly controlling signal event rates and kinematics in the context of long-lived particles that decay inside a neutrino detector. We demonstrate that the use of these simplified frameworks can not only allow for individual experimental analyses to be applicable to a wide variety of specific model scenarios but also serve as a bridge to model identification in the hopeful detection of a new physics signal. In the second direction, we develop theoretical formalism, construct extensions of the Standard Model, and explore their associated phenomenology in the cases of axion mixing with neutral pion in radiation bath, neutrino mass from the electroweak scale, and generalization of the N -Naturalness solution to the electroweak hierarchy problem.

Abstract: Two-dimensional electron gases (2DEGs) at complex oxide interfaces have emerged as a versatile platform for exploring novel quantum transport phenomena. Unlike conventional semiconductor 2DEGs, interfaces such as LaAlO$_3$/SrTiO$_3$ (LAO/STO) and LaAlO$_3$/KTaO$_3$ (LAO/KTO) combine gate-tunable electronic transport with intrinsic superconductivity, offering unique opportunities for investigating mesoscopic quantum devices.

In this talk, I will present these oxide interfaces as a versatile platform for hybrid quantum transport, highlighting their properties through acoustic, microwave, thermoelectric, and magnetotransport measurements. I will first discuss the generation and characterization of surface acoustic waves in LAO/STO and LAO/KTO. I demonstrate acoustoelectric transport in LAO/KTO, which can be used to shuttle charge carriers across quantum devices. I will then explore superconducting microwave resonators fabricated on LAO/KTO, highlighting their potential for parametric amplification. Next, I will present thermoelectric measurements of the superconducting state through the Nernst effect, providing new insights into superconductivity in LAO/KTO. Finally, I will discuss magnetotransport in coupled nanowire arrays fabricated in LAO/STO, illustrating the rich quantum transport phenomena that emerge in confined oxide nanostructures.

ABSTRACT:

The pancreatic islet is a heterogeneous micro-organ whose cellular composition and signaling vary across cell types, anatomical location, sex, and disease state. This dissertation develops complementary strategies to resolve that heterogeneity, first in human type 1 diabetes (T1D) and then in a mouse model of β-cell dopamine signaling.

In the first study, an AI-guided whole-slide imaging pipeline coupled to a hierarchical Bayesian and mixed-effects statistical framework was applied to immunohistochemically stained pancreata from 106 non-diabetic and T1D donors, quantifying all four endocrine cell types across more than two million islets. T1D produced a coordinated remodeling of islet composition: profound β-cell loss was accompanied by reciprocal α-cell expansion, whereas δ- and PP-cells were largely resilient. These changes were regionally concentrated, most pronounced in the pancreatic head, and were modulated by sex, age, and disease duration, with pseudotime analysis resolving distinct trajectories of compositional change across disease progression.

The second study examined the contributions of β-cell dopamine D2 receptor (D2R) signaling to glycemic control using a tamoxifen-inducible, β-cell-specific D2R conditional knockout (cKO) mouse model. Loss of β-cell D2R produced sex-dependent metabolic phenotypes: females showed blunted glucose-stimulated insulin secretion and diet-dependent glucose intolerance, whereas males showed enhanced glycemic control and islet secretion. Multiplex RNAscope revealed that islet D2R is expressed in a subpopulation of β-cells, suggesting that D2R may contribute to the emerging understanding of β-cell heterogeneity.

Taken together, both studies highlight important roles for sex differences in β-cell function. Just as importantly, this work uses complementary imaging, genetic, and physiologic approaches to define distinct populations of endocrine cell types, both under physiologic and disease contexts.

Abstract: Superconducting circuits are a leading platform for quantum information processing because of their flexibility as engineered quantum systems. Parametric controls provide an effective and flexible approach for manipulating superconducting circuits. In parametric control, appropriate choices of drive frequency, amplitude, and phase activate nonlinear Hamiltonian terms that turn on selected interactions and control them dynamically. In designing a SNAIL-mediated four-qubit quantum module, this work demonstrates parametrically activated single- and two-qubit control. However, design tradeoffs appear when stronger coupling and faster gates are pursued. To push parametric control further, this thesis uses transmons to explore subharmonic driving, subharmonic measurement, and a general strong-drive limit that applies not only to transmons but also to other superconducting circuits.

Subharmonic single-qubit control is demonstrated using a drive near one third of the qubit transition frequency, where the Josephson nonlinearity converts three drive photons into one excitation of the computational transition. This frequency separation allows the control line to be filtered strongly at the qubit frequency while still passing the lower-frequency control tone. With phase tracking, chirped modulation, transfer-function correction, and leakage-aware pulse tuning, this protocol realizes single-qubit gates with a best fidelity above 99.9% and π and π/2 gate times of 50.9ns and 37.4ns, respectively. I then extend the same idea to measurement by using an alternate subharmonic drive to generate a resonator field through the transmon nonlinearity. The resulting dispersive readout achieved a best measurement fidelity of 95.0%, and the model indicates that improved lifetime, frequency planning, and readout parameters can push the protocol toward much higher fidelity.

The second part of the dissertation studies the strong-drive threshold that limits parametric control. In experiments on strongly driven circuits, the driven mode population spreads into many highly excited transmon levels above a critical drive amplitude, causing the intended coherent response to disappear. I identify this breakdown as a speed limit for parametric interactions. By comparing simulations with experimental characterization, the model successfully predicts this threshold in driven transmons. This threshold model provides a tool for estimating the limits of parametrically driven systems and designing new devices that can further improve parametric control

Abstract
Prenatal genetic testing evaluates the presence of fetal genetic anomalies and is an essential part of evidence-based prenatal care. Prenatal genetic counselors (GCs) often facilitate testing and care delivery, allowing pregnant people to make decisions about their current and future pregnancies. Though prenatal genetic testing is recommended during prenatal care, it is costly and often inaccessible to low-resourced and uninsured pregnant people.

Medicaid coverage improves access to prenatal genetic services for low-income pregnant people, which may affect infant health outcomes. However, Medicaid coverage often varies across jurisdictions, and access to genetic services may be complicated by reproductive health policy changes following Dobbs v Jackson Women’s Health Organization (2022). This dissertation utilizes a multiple methods approach to better understand the policy landscape for prenatal genetic services and how it influences care delivery and infant health outcomes.

Aim 1 uses a rigorous legal mapping technique to characterize Medicaid coverage for non-invasive prenatal testing (NIPT), amniocentesis, and chorionic villus sampling in 51 United States jurisdictions as of 2025. We redundantly coded state-level policies, noting policy sources, effective dates, eligibility criteria, covered conditions, and prior authorization requirements.

Aim 2 uses national Pregnancy Risk Assessment Monitoring System data between 2011 and 2022 and a two-way fixed effects difference-in-differences analysis to estimate the impact of state Medicaid coverage for NIPT on birth defects. Our treatment group included those in states with NIPT coverage and information on birth defects. Our control group included those in states without NIPT coverage during our study. We controlled for individual-level and state-level factors affecting exposure to the policy and outcome.

Aim 3 consists of semi-structured interviews to understand how prenatal GCs operationalize practice-based competencies from their training in varying state policy environments. The interview guide included questions about day-to-day practice, a patient vignette, and if abortion policies affect care delivery. Interview transcripts were analyzed using deductive and inductive coding.

Our findings suggest that the policy environment affects prenatal genetic care delivery and infant health outcomes. This highlights the important role of policy in facilitating access to genetic services and underscores the potential implications of recent Medicaid and reproductive health policy changes.

Abstract: Over the last several decades, significant advances in our understanding of cellular and physiological processes that result in the initiation and progression of malignancy have been achieved. These insights have led to new treatments involving immunotherapy and targeted approaches to combat the cancer process. This presentation will discuss these advancements and take a look back at some of the individuals and their pioneering contributions that laid the foundation for the progress in cancer treatment that has been realized in recent decades. Finally, while the scientific and medical communities continue to pursue cures, the presentation will conclude by discussing how prevention, screening, and early intervention currently offer the best hope for achieving a cure.

Bio: Dr. Lomis is a fellowship trained surgical oncologist with extensive experience in the surgical management of complex cancer cases. He studied at the University of Pittsburgh, receiving a master’s degree in medicinal chemistry in 1987 and medical degree in 1992. He is the CEO and medical director of the San Fernando Valley Cancer Center and Valley Breast Care, with three sites in the Greater Los Angeles area. He conducts scientific research projects related to the development of novel compounds, including radioligand agents, to more precisely target cancer cells. He is the principal investigator for the San Fernando Valley site on several national clinical trials for Translational Research in Oncology-US Network. Current trials involve the evaluation of CDK4/6 inhibitors and antibody drug conjugates, one of which, the NATALEE trial, led to FDA approval of ribociclib for expanded use in the treatment of breast cancer.

Urban soils preserve long-term records of pollution produced by urbanization, industrial development, and atmospheric deposition. In historically industrial cities, these records are difficult to interpret because metal concentrations reflect overlapping effects of pollution sources, geology, climate, topography, soil chemistry, and atmospheric transport. This dissertation investigates metal pollution dynamics in complex urban environments by integrating soil geochemistry, sequential extraction, Pb isotope source tracing, atmospheric inversion analysis, gamma radiation records, unconventional well data, and HYSPLIT air-mass trajectory modeling.

First, I compared Pittsburgh soils with available New York City and Los Angeles datasets, showing that elevated Mn in Pittsburgh reflects industrial legacy, vegetation cycling, and metal retention processes. In contrast, Los Angeles showed greater exchangeable metal fractions, highlighting the role of climate in metal bioavailability.

Second, I investigated soil metal enrichment around the Clairton Coke Works. Soils surrounding the facility showed metal enrichment near the plant, although spatial patterns were not explained by distance alone. Pb isotope ratios indicated that soil Pb reflects a mixture of lithogenic background, legacy anthropogenic inputs, and a local coking-related industrial signature most evident near the facility and along atmospheric transport pathways.

Third, I analyzed long-term radiosonde, EPA RadNet gamma, unconventional gas well, and HYSPLIT trajectory datasets to examine how atmospheric conditions influence near-surface gamma radiation variability in Pittsburgh. These analyses show that atmospheric inversions influence gamma variability, especially under specific daytime, seasonal and air-mass direction conditions, while well-related signals appear indirect and dependent on transport pathways and boundary-layer stability.
Together, these findings show that urban pollutant patterns emerge from coupled interactions among soils, atmosphere, terrain, climate, and industrial history. Integrated geochemical and atmospheric approaches are therefore essential for identifying pollution sources, evaluating metal mobility, and interpreting contamination patterns in historically industrial regions.

ABSTRACT:

Retinogenesis requires precise synchronization of progenitor cell competence, spatial patterning, and cell fate specification. Retinal progenitor cells give rise to all retinal cell types in a highly ordered spatiotemporal manner. However, little is known about the gene regulatory mechanisms that maintain progenitor competence within retinogenesis. SOX2 is a key transcription factor expressed in retinal progenitor cells, a subset of amacrine cells, and Müller glia, and it is essential for neural competence. Mutations in SOX2 are associated with human eye developmental abnormalities, including microphthalmia and anophthalmia, underscoring its significance in eye development. However, the mechanisms by which SOX2 regulates retinal progenitor identity, patterning, and differentiation remain poorly defined.

I investigate the role of SOX2 during retinal development, focusing on how SOX2 regulates retinal progenitor cell identity, spatial tissue organization, and the transition from progenitor states to differentiated retinal cell types. This work investigates the effect of Sox2 loss in the developing retina and identifies regulatory changes associated with neural competence, spatial identity, and retinal cell fate specification by using genetic, molecular, transcriptomic, and chromatin accessibility approaches.

The findings show that SOX2 is required to sustain retinal progenitor cell identity and to support gene regulatory networks essential for retinal development and patterning. Loss of Sox2 disrupts progenitor identity by shifting the balance between progenitor maintenance and cell fate progression, altering retinal patterning, and affecting cell cycle progression. This work further identifies SOX2 as a key regulator of the boundary between the neurogenic neural retina and the non-neurogenic ciliary margin. Although this boundary is disrupted upon Sox2 loss, the core dorsoventral organization of the neural retina remains intact, suggesting that developmental competence and spatial patterning are controlled by partially separable regulatory programs. Overall, this dissertation identifies SOX2 as a central regulator of enhancer accessibility, neural retinal identity, progenitor competence, and spatial tissue organization during retinogenesis.

August's topic: A History of PM&R

Presented by: Elizabeth Sandel, MD

Elizabeth Sandel, MD has provided medical care to patients with brain injuries and leadership in her field for more than 40 years. She has practiced medicine and served in teaching, research, and administrative roles in healthcare systems on the East and West Coast, including the Jefferson and University of Pennsylvania Health Systems and Northern California Kaiser Permanente. She was a member of the Brain Injury Medicine Examination Committee of the American Board of PM&R from 2011 to 2015 and was one of the first group of physicians certified in that medical subspecialty in 2014. Now in retirement from clinical practice and research, Dr. Sandel spends much of her time educating the public about brain injury through her website and publications. Her book, Shaken Brain: The Science, Care, and Treatment of Concussion (Harvard University Press, 2020), is a resource for patients, families, and others who want to understand injuries to the brain and what interventions can help.

Abstract: As whole exome and genome sequencing become more prevalent, hundreds of genes associated with neurodevelopmental disorders (NDDs) have been identified. Among these are numerous genes encoding synaptic proteins that can have highly penetrant rare variants that are pathogenic. Our lab focuses on two families of postsynaptic proteins, NMDA receptors and neuroligins, that are high confidence NDD genes. We explore the synaptic consequences of these pathogenic de novo mutations. GRIN genes encode NMDA receptors subunits, which are glutamate receptors that are essential for neuronal development, learning, and memory. Endogenous NMDA receptors are typically composed of two GluN1 subunits and two GluN2 subunits. Our recent findings identify a cluster of pathogenic variants that lead to disruption of the PDZ binding site in GluN2B and protein destabilization. In other studies, our group is making mouse models to study pathogenic truncating mutations in GRIN2B and GRIN2A to determine the message stability, protein expression, and synaptic localization. By revealing the mechanistic consequences of genetic variants of NMDAR subunits, we hope to shed light on the mechanisms of disease and develop better therapeutics. Similarly, we are studying the effects of pathogenic rare variants on neuroligin function. We have identified multiple clusters of highly penetrant variants in NLGN4X that lead to protein trafficking deficits. In collaborations with National Center for Advancing Translational Sciences (NCATS), we have conducted drug screens and have several candidate molecules for rescuing the trafficking deficits in NLGN4X variants. We hope our discoveries can facilitate development of therapeutics for NDD disorders.

Offered by the Center for Mentoring in partnership with the Office of the Vice Provost for Graduate StudiesThis program introduces graduate faculty to the principles and best practices of mentoring graduate students, one-on–one and within mentoring programs or networks. Topics include:

1. Building student-centered mentoring relationships2. Establishing clear communication, expectations, and goals3. Fostering inclusivity and equity in mentoring4. Focusing on Individual Development Plans (IDP), often required by federal agencies (NSF, NIH)5. Practicing ongoing self-reflection to strengthen mentoring

Format & Requirements1. Pre-work: online module + reflective paragraph2. Two one-hour sessions (Sept. 11 & 18)3. Capstone: One-page mentoring plan

Connection to Mentoring AcademyThis course covers the first of four competencies of Mentoring Academy: Session One, specifically focusing on graduate mentoring. Completing Foundations of Graduate Mentoring may substitute for Session One in the Mentoring Academy series.

By the end of this session, participants will be able to:
1. Identify and describe the value of graduate mentoring.
2. Develop student-centered mentoring practices and relationships

September's topic: Unlocking Circuits and Designing Cross-species tools for Pain Discovery and Therapeutics

Presented by: Rebecca Seal, PhD

Rebecca Seal is a Professor of Neurobiology and Otolaryngology in the University of Pittsburgh School of Medicine. Research in her lab aims to understand how the various forms of persistent pain, across diverse pathological conditions, traverse the spinal cord with the goal of targeting the spinal cord to treat chronic pain. The lab employs a range of approaches in both rodents and primates and is pioneering non-invasive gene therapy. Dr. Seal is a Rita Allen Foundation Scholar and a Brain and Behavior Foundation Scholar and is Chair of the Pain Research Forum at the International Association for the Study of Pain, an active member of the Society for Neuroscience, and a co-Director of the Pittsburgh Pain Consortium. She previously served on the organizing committee to create the US Association for the Study of Pain (USASP) and served on the Board of Directors of USASP.

Students who enroll and complete the series (including all participation and assignment-based requirements) will be awarded a Certificate of Completion and are eligible to join the next cohort of the Big Idea Center’s formal recognition opportunity – The ChangeMaker Scholar Program.

Who can join?

Any currently enrolled Pitt student (undergraduate through postdoc) of any discipline who wants to learn how to create and impact change.

What does it cost?

This program is free to Pitt students. The only “cost” is your time and effort!

All sessions will be held in-person.

The ChangeMaker Series will cover the following topics. Participants will receive a full schedule and speaker list once added to their cohort Canvas site. Session topics are subject to change.

Participants will discuss secondary traumatic stress (“STS”) and its relevance to these trends. We will define STS and two related problems, compassion fatigue and burnout. Then we will consider symptoms of these and how to develop resilience through a personal “compassion satisfaction plan” that protects their well-being and effectiveness in these settings.

Fostering Independence (Competency #6)Promoting Professional Development (Competency #7), and Promoting Professional Development and Articulating Your Mentoring Philosophy and Plan (Competency #8)

Sessions are conducted through interactive case studies. Mentoring Academy members, who are trained faculty recognized for their mentoring contributions at the University of Pittsburgh, will facilitate each session. Participants can attend one or all Mentoring Academy sessions to earn a credential covering all eight competencies, based on the curriculum from the Center for the Improvement of Mentored Experiences in Research.

Participants will be able to:

Define independence, its core elements, and how those elements change throughout a mentoring relationship.Employ strategies to build mentee confidence, establish trust, and foster independence.Identify the benefits and challenges of fostering independence.
